WP4 – Phenotyping and Omics analyses will quantify the phenotypes and physiological parameters (specific growth rate, biomass yield, etc.) of all the strains with reduced genomes to select the most appropriate strains for biotechnological application. Strain selection will be based on process criteria defined in WP5, and on discrepancies with model predictions. Selected strains will be characterized using quantitative metabolomics and fluxomics methods specifically adapted for B. subtilis, as well as quantitative proteomics, phosphoproteomics, and transcriptomics. In addition, GFP reporters fused to various selected promoters (Live cell array and time-lapse fluorescence microscopy) will be used to fine tune the operation of synthetic pathways and some strains.
